Reagent for deoxofluorination
Pyridine-2-sulfonyl fluoride (PyFluor) serves as a reliable reagent for the fluorination of alcohols, offering outstanding stability and a simple reaction procedure1. It represents an advancement over the traditional DAST reagent. Primarily PyFluor is designed to enhance selectivity against elimination reactions, facilitating efficient and straightforward purification processes. Unlike DAST, which often yields unsaturated side products and exhibits poor thermal stability, PyFluor surpasses such issues, resulting in higher reaction yields. PyFluor also tolerates a variety of basic functionalities including phthalimides, heterocycles, and even unprotected amines and anilines. Most reactions proceed at room temperature, although cyclic or sterically encumbered substrates require moderate heating2. The combination of selectivity, safety, and cost-effectiveness makes PyFluor an ideal choice for deoxyfluorination on a preparative scale.
