N-Methyl-5-phenyl-1,2,4-triazole sulfonyl fluoride

N-Methyl-5-phenyl-1,2,4-triazole sulfonyl fluoride (SF-3) is a highly reactive heteroaryl sulfonyl fluoride reagent developed as a next-generation deoxyfluorination reagent with improved reactivity, handling properties, and safety compared with PyFluor, PBSF, and DAST. Identified through a data science-guided reagent discovery workflow1, SF-3 combines a high melting point (128 °C) with exceptionally rapid deoxyfluorination kinetics, achieving near-complete conversion of model alcohol substrates within minutes while maintaining a favorable thermal safety profile for laboratory use. The reagent is effective for the deoxyfluorination of a broad range of alcohols, including primary and secondary aliphatic alcohols, strained cyclic alcohols, activated benzylic alcohols, natural products, pharmaceutical intermediates, and drug-like molecules. SF-3 is particularly advantageous for challenging cyclic alcohols, substantially outperforming PyFluor and enabling efficient late-stage fluorination of complex substrates, including reserpine and dihydroartemisinin derivatives, under relatively mild conditions.
