Reagent for peptide synthesis, Barton esterification

HOTT

HOTT is a reagent used to convert carboxylic acids into Barton esters, facilitating reductive and oxidative decarboxylation, peptide coupling, and amidation1. This white solid reagent should be protected from light due to the sensitivity of 2-mercaptopyridine N-oxide. HOTT is an efficient and easy-to-handle reagent that simplifies work-up processes in various organic synthesis applications. In the case of peptide synthesis, the reaction conditions are very mild and do not adversely affect other functionalities prone to nucleophilic attack. A simple extractive work-up is sufficient to obtain the primary amides in pure form.
