Reagent for trimethylsilylation
N,O-Bis(trimethylsilyl)trifluoroacetamide (BSTFA) is a widely used reagent for trimethylsilylation1. It converts active hydrogen-containing groups (such as acids, alcohols, thiols, and amines) into volatile and thermally stable trimethylsilyl derivatives suitable for gas chromatography (GC) analysis. BSTFA is effective for in situ trimethylsilylation in chemical transformations, enabling the formation of nucleosides and N-glycosidic bonds, and palladium-catalyzed asymmetric allylic alkylation. The reagent is a colorless to pale yellow liquid highly sensitive to moisture, requiring storage in a cool, dry environment. It should be handled with proper ventilation and protective equipment due to its volatility and sensitivity to moisture.
