Reagent for construction of meta-fluoropyridines

(2Z)-3-(diethylamino)-2-fluoroprop-2-enal

(2Z)-3-(Diethylamino)-2-fluoroprop-2-enal is a neutral, shelf-stable fluorinated enamine reagent developed as a safe alternative to pyridyl diazonium salts for accessing meta-fluorinated pyridines1. The reagent acts as a modular C–C–N synthon that undergoes condensation–annulation with bis-nucleophilic amines (amides, amidines, enamines, amino-heterocycles) in the presence of Me₃SiCl as a condensing agent. Under mild heating in pyridine (≈85 °C), it enables the direct construction of meta-fluoropyridine rings in a single step. The reagent shows broad substrate scope, tolerating electron-rich and moderately electron-poor heterocycles and delivering over 40 new meta-fluorinated pyridines across 15 heterocyclic classes.
