Reagent for synthesis of thiols
2,4,6-Trimethoxybenzyl mercaptan is a reagent used primarily for thiol synthesis1. It can serve as a protecting group for thiols in organic synthesis, forming 2,4,6-trimethoxybenzyl (TMB) thioethers that are stable under a wide range of conditions (acidic, basic, and soft oxidation/reduction conditions), yet can be cleaved selectively. The reagent offers mild cleavage conditions, often via oxidative or acidic treatment, enabling orthogonal strategies in multi-step synthesis. This stability makes it useful in later stages where the thiol functionality needs to be masked. The bulky and electron-rich 2,4,6-trimethoxybenzyl group also provides steric protection, reducing unwanted side reactions.
