Reagent for carboxylic acid activation
N-hydroxysuccinimide (HOSu) finds significant application in the formation of isolable activated derivatives of Nα-protected α-amino acids1. These derivatives then readily participate in smooth coupling reactions with amino esters. The typical method involves using 1,3-dicyclohexylcarbodiimide (DCC) as the initial coupling agent (HOSu-DCC method). Alternatively, N-hydroxyphthalimide (NHPI) can be used similarly. However, HOSu holds a distinct advantage due to its high water solubility. This allows easy separation and purification of the activated derivatives, making HOSu an advantageous choice for such coupling reactions. The HOSu–DCC method is not limited to peptide synthesis and is well suited to preparing amides from ammonia and primary amines.
