Small bicyclic amines often form strained molecular systems, leading to unusual properties and reactivity. For instance, derivatives of 1-azabicyclo[1.1.0]butane serve as emerging reagents for recently developed strain-release azetidine ring formation. Beyond their use as reactants, strained amines are valuable in biomolecular engineering, imparting bioactive molecules with unique geometric and dynamic features. Recently, Enamine synthesized the first molecule featuring a 5-azabicyclo[2.1.1]hexane scaffold, a long-anticipated, highly strained bicyclic amine expected to induce the highest level of amide pyramidalization.
