Reagent for protection

Triphenylmethanesulfenyl chloride (TrSCl) is a reagent used for introducing oxidatively cleavable protection on the amino group1. TrSCl can also be used for the protection of the 5′‐hydroxy group in deoxynucleosides. It forms sulfenamide and sulfenate linkages that serve as the basis for TAB and MOB protecting groups. These groups enable acid-free DNA synthesis, as they can be removed under mild oxidative conditions (e.g., aqueous iodine). The reagent's stability and selective reactivity make it valuable for nucleoside and oligonucleotide modification workflows.
