π-Stacking is one of the major non-covalent interactions involved in drug affinity to their protein targets. Introduction of heteroaromatic residues allows fine tuning on the π-stacking with aromatic residues. In addition, heteroatoms introduce previously non-existent hydrogen bond sites in the structures.
